He was encouraged by his family to pursue his musical talents, so he learned the ukulele and guitar at a young age. Leo was performing professionally by the time he was thirteen. Initially, he was drawn to jazz because of his inspiration from jazz guitarists like Charlie Christian, Barney Kessel, Johnny Smith and Kenny Burrell. He has also made a statement on the rhythm and blues and funk genres as well.  When he was fourteen, he was playing back-up for Otis Redding and Clyde McPhatter. After serving in the army at seventeen for two years, he joined Art Neville and the Neville Sounds and began forming his band, The Meters.

The band released two hit singles, Sophisticated Cissy and Cissy Strut, as well as released eight studio albums from 1969 to 1977. Leo Nocentelli wrote most of the music that the band released, and he won a Grammy Lifetime Achievement Award as a member of The Meters. After his time in The Meters, he moved to Los Angeles in 1982 and performed in several reunions with the original Meters band. He moved to New Orleans in 2015 and continues to perform in various bands of jazz and funk.
